Evidence that toxin resistance in poison birds and frogs is not rooted in sodium channel mutations and may rely on “toxin sponge” proteins

چکیده

Many poisonous organisms carry small-molecule toxins that alter voltage-gated sodium channel (NaV) function. Among these, batrachotoxin (BTX) from Pitohui poison birds and Phyllobates frogs stands out because of its lethality unusual effects on NaV How these toxin-bearing avoid autointoxication remains poorly understood. In frogs, a DIVS6 pore-forming helix N-to-T mutation has been proposed as the BTX resistance mechanism. Here, we show this variant is absent frog NaVs, incurs strong cost compromising function, fails to produce BTX-resistant channels in NaVs.

Convergent Substitutions in a Sodium Channel Suggest Multiple Origins of Toxin Resistance in Poison Frogs.

Complex phenotypes typically have a correspondingly multifaceted genetic component. However, the genotype-phenotype association between chemical defense and resistance is often simple: genetic changes in the binding site of a toxin alter how it affects its target. Some toxic organisms, such as poison frogs (Anura: Dendrobatidae), have defensive alkaloids that disrupt the function of ion channel...

Evidence that Lipopolysaccharide and Pertussis Toxin Bind

In previous studies, we used a photoactivable, radioiodinated lipopolysaccharide (LPS) derivative to define and characterize a specific bacterial endotoxic LPS-binding protein (p73) on mammalian lymphoreticular cells, including B and T lymphocytes and macrophages. More recently, using the same methodology, we characterized a specific interaction of LPS with the S2 subunit ofBordeteUa pertussis ...

Botulinum toxin the poison that heals: A brief review

Botulinum neurotoxins, causative agents of botulism in humans, are produced by Clostridium botulinum, an anaerobic spore-former Gram-positive bacillus. Botulinum neurotoxin poses a major bioweapon threat because of its extreme potency and lethality; its ease of production, transport, and misuse; and the need for prolonged intensive care among affected persons. This paper aims at discussing botu...

BACKGROUND: Campylobacteriosis is one of the most commonbacterial intestinal disorders of humans in many countries.A wide range of domestic and wild bird species have been identifiedas natural reservoirs. OBJECTIVES: The aim of this studywas to determine the Campylobacter spp from pet birds andscreen the determined Campylobacters for presence of virulencecytolethal distending toxin (cdt) genes....
